Apolipoprotein E alleles have distinct effects on the rate of cognitive decline in Alzheimer’s disease
In a population with autopsy-proven Alzheimer’s disease, APOEε2 and APOEε4 alleles had divergent effects on the rate of cognitive decline.
Why this matters
This unique study employed a novel reverse longitudinal modeling approach that utilized data from individuals with confirmed Alzheimer’s disease; this method controlled neuropathological comorbidities that could affect progression rate, so that the focus remained on the effect of APOE genotype. The findings suggest that, besides neuropathology, other apolipoprotein E-related phenotypes drive clinical progression in Alzheimer’s disease.
