Insulin resistance may have a genetically based causal role in small vessel stroke and Alzheimer’s disease (AD).
Why this matters
Cerebral small vascular disease (CSVD) and AD are common pathologies with overlapping risk factors that include hypertension and altered glucose metabolism.
Insulin resistance has been associated with stroke, CSVD, and AD, but evidence for a causal role in previous Mendelian randomization studies – a study design that simulates a randomized control trial – has been mixed.
Further investigation of insulin resistance in CSVD and AD using a Mendelian randomization approach is warranted.
